DanioScope 1 - Morphology and Analysis

Morphology
Aim: To measure body parts and organs. Typical measurements are body length, eye area, and pericardial area.
Procedure:
- Choose Acquisition > Morphology and click Add Images or Add Images From Video to load your images.
- Use a Calibration profile to apply the same scale to multiple images. To create a new Calibration profile, click the Calibration Profiles button. Click Add and choose a picture containing an object of known size, for instance a stage micrometer. This picture must be obtained with the same lens and zoom level as the pictures of the embryos. Next, draw a line that connects the two extremes of the object, enter a value for Known distance and click OK.
- Select the Calibration profile you want to use.
- Draw the measurement line/shape for Body Length, Eye Size or Pericardial Area. For other measurements like the number of somites, enter or select the appropriate value.

Calculate Statistics
Trial Statistics
Choose Analysis > Trial Statistics to get statistics per subject. You can calculate statistics like Burst Activity and Burst Count for Activity, Beats per second or Beats per minute for Cardiology.
Group Statistics
Choose Analysis > Group Statistics and Charts to calculate average values of the statistics per group. Define groups based on the values of the independent variables in your experiment.
Click the Comparison Settings button to compare different types of measurements, for example, Burst Count with BPM (beats per minute). Click the Comparison tab to view the corresponding statistics and charts.
Other Options
- Choose Analysis > Data Selection to filter data based on the values of the independent variables. For example, to analyze only subjects with Time point = 48 hpf. Select the check box under Filter for the independent variable of your choice.
- Choose Analysis > Visualizations to view activity plots for the subjects in your videos, the activity in the heart area or the flow in an artery or gut.

Export Data and Results
To Export the Trial Statistics or the Group Statistics
Choose the option you require under Analysis. Click the Export Statistics button. Select the export format and click Export.
To Export the Raw Activity Data
For Activity, Cardiology and Flow you can also export the raw data recorded at each video frame. In the Trial Statistics screen, click the Export Raw Data button.
To Back Up an Entire Experiment
Use the backup function to keep your data safe in another location. Make a backup of your experiments on a regular basis, at least once a day.
- Choose File > Make Backup. Give the backup file a name and choose a destination.
- Copy the backup file (with extension
*.dsz) to the other computer, together with the video and image files. - To restore an experiment from a backup file and open it with DanioScope on the other computer, choose File > Restore Backup. Choose the backup file and select the folder where you want to store the experiment.
