Add a Measurement

Prerequisites

Choose the option you require from the Acquisition menu.

To Add a Measurement

1. At the top of the video window, choose a drawing tool.
2. Draw a shape around the body part of interest. To enlarge the image, use the Zoom In button. When drawing a polygon, click once to add a new vertex. When ready, release the mouse button (for a rectangle or oval) or double-click the first vertex (for a polygon).

The next steps depend on whether subjects are already defined.

To Add a Measurement When No Subject Is Defined Yet

1. If no subject is defined yet for that video/image, the Add Subjects window appears. Enter the Number of subjects to measure in the video/image. The new area of interest will be assigned to S1/M1. If the video/image contains one subject, leave 1.
2. Optionally, select the Independent Variable Values for this group of subjects. If subjects must be assigned to different values (e.g. some Control and some Concentrate 1), do this in the list under the video window.
3. Select Draw template shapes for all subjects to create multiple copies of the shape you have just drawn. You can later move and edit each shape to adapt it to the corresponding area of interest. If you leave the option not selected, DanioScope adds the subjects to your list but does not create more shapes.
4. Click Add.

To Add a Measurement When One or More Subjects Are Already Defined

1. A context menu appears, listing the subject names. If the new area of interest refers to a subject already defined, select that subject. The new area will be marked as a new measurement for that subject (for example, S01/M2). If the new area of interest refers to a new, undefined subject, select Add subject from the context menu.
2. Enter 1 or a higher number, depending on how many subjects are still undefined, and optionally specify the values of the independent variables for those subjects (you can also do that later on). Next, click Add. The shape is marked as a new measurement for the first of the added subjects (for example, S04/M1).

To Adjust the Area of Interest

If necessary, drag the shape handles to move and resize it so that it matches the contour of the subject/body part as much as possible. For Activity, a little overlap between the circles is acceptable.

Important: If you resize a shape after acquisition, the statistics for that measurement are cleared and the Status column says Next: Acquire. Click Acquire to acquire the data.

Delete a Shape (Measurement or Subject)

Do one of the following:

Delete a Shape on the Video Image

Right-click a shape and select Delete Measurement. Select Delete Subject if you want to delete the subject the shape belongs to. This means that all measurements and shapes for that subject are removed. Deleting a shape also means removing the measurement in the list. The subject is deleted from the list only if you choose Delete Subject.

Delete a Row in the Measurement List

Right-click the row in the list under the video window and select:

• Delete Measurement. The measurement row is deleted. If that row is the only one for a subject, it is cleared (the cells Measurement Start, Stop and Duration are blank), but not deleted.
• Delete Subject. All rows for that subject are deleted.

The shapes are removed from the video window according to the option chosen.

To select multiple shapes/measurement rows, hold the Ctrl key down and click each shape. To select all measurement rows, click the top row, hold down the Shift key and click the last row.

Add a New Subject

You add a subject by drawing a new shape and selecting Add Subjects. See Add a Measurement.

Edit the Independent Variable Values

Aim

To assign values of independent variables like Time Point and Compound to different subjects and measurements. You can then use those values to group subjects and measurements in Group Statistics and charts. If subjects in the same video have different values of an independent variable, you can set those values for each subject.

To Assign a Value to One Subject/Measurement

Do one of the following:

• In the measurement list under the video window, locate the independent variable column. Select the appropriate value for a subject.
• In the video window, right-click the subject/measurement you want to edit and choose Edit Variable Values. In the window that appears, select the appropriate value for that subject/measurement.

To Assign a Value to Multiple Subjects/Measurements

Do one of the following:

• In the video window, hold Ctrl or Shift down and click each subject/measurement you want to edit. When ready, choose Edit Variable Values. In the window that appears, choose the appropriate value and click OK.
• In the measurement list under the video window, select the rows for subjects and measurements you want to edit. To select adjacent rows, hold Shift down and click the first and the last row. To select non-adjacent rows, hold Ctrl down and click each row. Next, right-click anywhere in the rows and select Edit Variable Values. In the window that appears, choose the appropriate value and click OK.

For both methods, the value of the independent variable chosen is applied to all subjects/measurements selected.

Duplicate a Shape

To make an exact duplicate of a shape (recommended when comparing Flow measurements), right-click a shape and select Duplicate. In the context menu that appears, select an existing subject to assign the shape to it or select Add Subjects. If you select an existing subject with n measurements, the shape receives the same subject code and a new measurement code (n+1).

Edit a Shape

• To move one of the ends of a line, select the line and drag the corresponding white square.
• To reposition the vertex of a polygon or open polyline, hover the mouse on the shape; when a four-arrows cursor appears, click once. Drag the white square at the vertex you require.
• To move an entire line or other shape, click in the middle of the line or shape and drag it to the new position.
