Y-Maze Experiment Setup in EthoVision XT – Protocol and Zone Configuration Guidance

Last updated: Nov 01, 2026

Problem

Users new to EthoVision XT need guidance on setting up a Y-maze experiment, including arena zone definition, trial control, and analysis parameters for spontaneous alternation or other Y-maze protocols.

Details

New EthoVision users or researchers unfamiliar with maze-based setups in EthoVision XT ask for step-by-step protocol guidance for the Y-maze task. The Y-maze involves three arms that must be defined as distinct zones, and alternation scoring requires tracking sequence of zone entries. Relevant for EthoVision XT across versions.

Solution

1. Refer to the official EthoVision XT Y-maze protocol document available from Noldus support or the MyNoldus knowledge base; contact support to request the protocol manual and any available training recordings.
2. In Arena Settings, draw three separate zones corresponding to the three arms of the Y-maze. Name them clearly (e.g., Arm A, Arm B, Arm C).
3. Define the center of the maze as a separate zone if you wish to exclude it from arm entry counts, or leave it as the background zone.
4. Use the 'In zone' parameter and zone entry counts to track how often the animal enters each arm.
5. For spontaneous alternation, use a sequence analysis or conditional variable approach: define a variable that tracks the last two arms visited and flags an alternation when all three arms are visited in sequence without repetition.
6. Set trial duration in Trial Control to match your standard Y-maze exposure time (e.g., 5 or 8 minutes).
7. Calibrate the arena using a known reference distance before acquisition.
8. Export zone entry sequence data and calculate alternation percentage in your analysis software (e.g., Excel) or using EthoVision's built-in sequence analysis if available in your licensed version.
